Overview

[212Pb]Pb-AG1206 is an investigational targeted alpha therapy (TAT) radiopharmaceutical consisting of the alpha-emitting radioisotope Lead-212 ([212Pb]) conjugated to AG1206, a small molecule ligand with high affinity for Prostate-Specific Membrane Antigen (PSMA). Developed by AlphaGen, this agent is designed to treat metastatic castration-resistant prostate cancer (mCRPC) by delivering high-linear energy transfer (LET) alpha radiation directly to PSMA-expressing tumor cells. Lead-212 serves as an in vivo generator for alpha-emitting progeny, including Bismuth-212 and Polonium-212, which induce lethal double-strand DNA breaks that are difficult for cancer cells to repair. This approach aims to overcome the limitations and resistance associated with beta-emitting radiopharmaceuticals, such as Lutetium-177 based therapies, by providing more potent localized cytotoxicity with minimal damage to surrounding healthy tissue.
